Construction of the franchise in the car park near the new Victoria Point bus station will begin on Monday.
The outlet with a dual lane drive-through and dine-in facilities is expected to open around late June.
Franchisees German Leudo, his wife Veronica Junca and brothers David Leudo and Sergio Garcia, from Colombia, said they would employ about 30 people.
German Leudo, who works at Capalaba's Guzman y Gomez which opened in December, said they were excited to launch at Victoria Point.
Some car parks near the former Blockbuster store will be lost but others would be created near the Victoria Point bus station which is being built on Cleveland-Redland Bay Road.
Mr Leudo said the restaurant would go along with the Guzman y Gomez tradition of handing out free burritos on the day they opened.
He said they looked forward to being part of the Victoria Point community and would look for opportunities such as sponsorship of sports teams.
Victoria Point Shopping Centre manager Jamie Sneyd said centre owner Leda Group was pleased to be involved in the development.
"Leda Group is constantly exploring opportunities for further development and expansion in order to meet the needs of our growing local community," Mr Sneyd said.
"Guzman y Gomez marks the first of several improvements happening at Victoria Point Shopping Centre now and in the near future."
